Detection of PME activity Pectin methylesterase activity was measured using an ELISA kit (Shanghai Hengyuan Biological Technology Co., Ltd). Approximately 0.1 g fresh weight of root tips (0–10 mm) was homogenized in 0.9 mL of PBS (50 mM, pH 7.0), and the extracts were centrifuged at 12,000g at 4 ℃ for 20 min. The crude enzyme extracts were used to determine the activities of pectin methylesterase by following the manufacturer’s instructions.
